父系小鼠高甲基化饮食对子代小鼠学习记忆的影响与机制研究

  DNA甲基化(DNA methylation)是指在甲基转移酶(DNA methyltransferases,DNMT)的催化下,在基因组CpG二核苷酸的胞嘧啶5’碳位共价键结合一个甲基基团,形成5-甲基胞嘧啶。DNA甲基化修饰改变了DNA的物理结构,阻止DNA结合蛋白在转录过程与DNA链的结合,从而调控基因表达,是重要的表观遗传调节方式之一。DNA甲基化对营养高度敏感,食物中的叶酸(folic acid,FA)、维生素B12、甜菜碱(betaine)、甲硫氨酸(methionine)和胆碱(choline)等成分是重要的甲基供体,通过一碳单位代谢途径直接或间接影响DNA甲基化。大量的流行病学统计数据和动物研究表明围孕期甲基供体营养素的不足或过剩,可通过影响DNA甲基化等表观遗传修饰,改变部分关键基因的表达,从而引起子代多种神经发育、行为和神经认知功能等表型的改变。但上述的研究几乎都集中在母体因素,缺乏对父系甲基供体摄取对子代影响的考察。已有研究证据表明,精子细胞的表观基因组会受到父亲环境暴露因素的影响,从而引发子代表型的改变,所以为了探讨父亲高甲基化饮食暴露对子...
